Compounds (a) ( 3.00g, 8.0mmol ) and 50mL 30% hydrogen peroxide in 800mL acetic acid, heating 3.5h in 70^ 80 . In 45^ 55  concentrate the reaction to the 1/3 volume, dumped 3L ice into the remaining material. Filter collection precipitation, after crystallization of purified get 2.4G orange needle crystal 7- ethyl camptothecin -1- oxygen (b), the yield is 78%, melting point is 255 .

Compounds (b) ( 1.00G, 2.6mmol ) and 2.6mLlmol/L sulfuric acid dissolved in 1L two oxygen six alkanes, nitrogen bubbling air for 20min. Under stirring, with Pyrex filter of high pressure mercury lamp ( 450W, Usio UM-452 ) lighting 30min. The reaction liquid concentrated to dry, the remainder dissolved into 50mL 10% methanol chloroform. The solution washed with the 500mL water, both organic and inorganic layers through a celite ( Celite ) column, two-phase insoluble in all adsorbed thereon, and then with 10% methanol chloroform ( 3 × 200mL ) elution. Concentrating the eluent, after residue washing with methanol recrystallization, get 0.5g pale yellow crystals 7- ethyl -10- hydroxy camptothecin, the yield is 49%, melting point is 216 .

Compounds (c) ( 5.00g, 12.8mmol ) dissolved in 10 L two oxygen ring six and 50mL three ethylamine, at room temperature and stirring, pour into phosgene (produced by 3.75mL diphosgene acting on the activated carbon), continue to stir for 1h. Filtration, filtrate decompression and concentration to dry. Residue impregnated with acetone then filter, get 5.2g powder colorless compounds (d), the yield is 90%.

Compounds (d) ( 3.0g ) soluble in dichloromethane and methanol ( 500mL-150mL ) and 15mL pyridine mixture, under stirring, dropping double compounds (e) of dichloromethane solution. Then continue to stir for 15h at room temperature. Decompression and concentration to dry, residual soluble in dichloromethane, after washing with 7% sodium bicarbonate solution, anhydrous magnesium sulfate drying. Filter, vacuum evaporate. The remainder with silica gel column chromatography, 4% methanol-dichloromethane elution. Get light yellow powder products, the yield is 79.8%, melting point is 222 ~ 223 .

Irinotecan for adult metastatic colorectal cancer treating, for containing 5 Fu chemotherapy failure patients, it can be used as second-line treatment. At the same time,irinotecan price is reasonable, application in a variety of clinical trials of gastric cancer, esophageal cancer, extensive stage small cell lung cancer are conducting, it has come to the stage of observation results, has good clinical application prospect, worthy of close attention.
